Now, for the first time, a team led by scientists at The Scripps Research Institute and Sanford-Burnham Medical Research Institute has determined the 3D structure of the interaction between this critical bacterial protein and an immune molecule called TLR5, shedding light on how the body protects itself from such foreign invaders.

The study, published February 17 in Science, not only helps decipher the molecular mechanism underlying TLR5 recognition and function, but it also advances knowledge that?s key to the design of new therapeutics.

?The structure of the TLR5-flagellin complex visualizes molecular events that occur on the cell surface to trigger flagellin-induced host immune responses, and provides significant insights into the structural basis for TLR5 recognition and signaling,? said Ian Wilson, D.Sc., Hansen Professor of Structural Biology at Scripps Research who led the study with Andrei Osterman, Ph.D., professor in Sanford-Burnham?s Infectious and Inflammatory Disease Center.

?Gaining knowledge of a molecular interaction and action?as we did in this study? is critically important to the further development of therapeutics based on agonists and antagonists of the TLR5 receptor,? said Osterman.

Flagellin is a component in some vaccines and a derivative of this protein is currently being developed as a medical countermeasure to radiation by Cleveland BioLabs, Inc. (NASDAQ:CBLI), also a contributor to the new study.

Keeping an eye out for infection

Some of the body?s first lines of defense against invading bacteria are Toll-like receptors (TLRs), sensors that sit on the surface of many different types of cells. There are roughly a dozen different TLRs, each keeping an eye out for a particular sign of infection.

TLR5, for example, specifically recognizes and binds to flagellin. Like most TLRs, TLR5 does more than just sense bacteria?it also sends signals that call up immune cells to destroy the intruder. But to fully understand how TLR5 works, scientists needed to be able to see its 3D shape and how it binds to flagellin.

The structures of several other TLRs had already been solved, but each of these binds non-protein molecules, such as RNA or lipids. For technical reasons, determining the structure of TLR5?the only TLR that binds a protein?had long been a challenge.

In this study, the Scripps Research team was able to overcome these hurdles using TLR5 found in zebrafish as a proxy for the human protein. The scientists were then able to apply a technique called X-ray crystallography, which uses powerful X-ray beams to produce 3D images of proteins at the atomic level.

At Sanford-Burnham, Osterman and his team used biochemical and protein engineering methods to unravel the mechanistic details of interactions between TLR5 and flagellin and its derivatives.

Scientists at Roswell Park Cancer Institute and Cleveland BioLabs, Inc. in Buffalo, under the leadership of Andrei Gudkov, Ph.D., performed complementary experiments in human cells expressing TLR5 and validated the fish TLR5 as a good surrogate for human TLR5.

This research was funded by the National Institute of Allergy and Infectious Diseases, the Skaggs Institute for Chemical Biology at Scripps Research, and Cleveland BioLabs, Inc. In addition to Wilson, Osterman, and Gudkov, the study?s co-authors include Sung-il Yoon, Scripps Research; Oleg Kurnasov, Sanford-Burnham; Venkatesh Natarajan, Roswell Park Cancer Institute; and Minsun Hong, Scripps Research.

SAN FRANCISCO (Reuters) ? Guests at some New York Fashion Week shows and others watching online now have a new way to identify the looks they see sashaying down the runways.

The app called Made Fashion Week is companion app for people attending the shows, delivering instant content including images of models, item information and designer biographies in sync with what is seen on the catwalk.

"The moment that a model appears on the runway, there's audio sent through the public address system, delivering the photo that is snapped when she gets three feet (one meter) out of the actual curtain," said Jonathan Glanz, the CEO of Sonic Notify, which developed the technology.

"The turnaround from the time the model hits the runway to the time the photo turns up on a device is three seconds."

It also allows attendees to take notes on items, create custom lookbooks and to share favorite designs on Twitter.

Glanz said the aim of the app is to serve as a communication channel for designers to engage viewers during the show, but it can also be used beyond the runway, for example to notify a shopper as to when an item has finally hit stores.

"Because you were at the show, your phone will go off four months from now saying you can go buy these clothes today at Barney's or Macy's," he said.

The content is streamed through high-frequency audio signals that are delivered along with the audible sound one would hear at an event. The technology is plugged into a venue's mixing boards, and content is encoded and transmitted to mobile devices without relying on GPS or WiFi.

"We're a passive solution for delivering instant content that's in the actual sound coming out," he explained.

Glanz said that Sonic Notify is working with other companies and musicians to create more engagement during live events by sending multimedia content, like songs, coupons or advertisements, to mobile devices.

"A lot of live events have become manufactured. It feels like they're coming out of a printing press," he said.

He believes that this type of presence-based content will become more mainstream at live events, where attendees are looking for more engagement.

For example, the technology might be used at a concert to trigger voting for the encore song.

"You really create the interaction that nothing can provide today in that exact timed, orchestrated way. It just engages at a whole different level," he said.

Excluding one-time items in the latest period, Goodyear said that its earnings were 3 cents per share. Analysts expected earnings of 20 cents per share, according to a FactSet survey.

The prior-year period included a $160 million charge tied to its plans to close a plant in Union City, Tenn.

Quarterly revenue increased 12 percent to $5.68 billion from $5.07 billion partly on higher tire prices, but tire unit volumes dropped 5 percent on difficulties in Latin America, flooding in Thailand and declining replacement industry volumes in mature markets.

The revenue performance missed Wall Street's estimate of $5.86 billion.

Goodyear said that revenue per tire rose 19 percent, taking out the effects of foreign currency translation.

Middle distance fields get even faster at USA Track & Field Classic
INDIANAPOLIS ? The fast fields competing in the Geoff Hollister men?s mile and women?s 1,500 at Saturday?s USA Track & Field Classic in Fayetteville, Ark., have gotten faster. Five newly announced entrants, including a World Indoor silver medalist and World Junior champion, deepen the fields.

With the addition of two-time World Indoor silver medalist Daniel Kipchirchir Komen of Kenya (3:48.28PR), the Geoff Hollister Mile now includes five men with personal bests of 3:50 or faster. Also added to the field is American Russell Brown (Stanford 2008), who ran a PR of 3:51.45 at the 2011 Nike Prefontaine Classic. Announced on Wednesday were World Outdoor silver medalist and U.S. Open champion Silas Kiplagat of Kenya (3:49.39PR), four-time NCAA mile and 1,500m champion Leo Manzano (3:50.64PR), 2010 5th Avenue Mile champion Amine Laalou of Morocco (3:50.22PR), and 2010 World Junior champion Caleb Ndiku of Kenya (3:49.77PR).

The women?s 1,500 now includes New Balance Indoor Grand Prix 1,000m champion and World Outdoor fourth-place finisher Btissam Lakhouad of Morocco (3:59.35PR), 18-year-old World Junior champion Tizita Bogale of Ethiopia and Siham Hilali of Morocco (4:01.33PR). They join a field with previously announced Americans Anna Pierce (3:59.38PR), three-time USA outdoor 1,500m champion Treniere Moser (4:03.32PR), and U.S. Open winner Brenda Martinez.

Held at the University of Arkansas? Randal Tyson Center, the USA Track & Field Classic is the third stop on USATF?s 2012 Indoor Visa Championship Series. Huddle, Braun head to St. Louis for USA Cross Country Championships
INDIANAPOLIS ? Molly Huddle (Providence, R.I.) and Aaron Braun (Flagstaff, Ariz.) lead the fields this Saturday as the city of St. Louis will host the first of two consecutive USA Cross Country Championships. Both the 2012 and 2013 editions will be held at Forest Park, the site of the 2004 Women?s Olympic Trials Marathon.

In addition to the national titles on the line in St. Louis, open and junior men and women will qualify to represent Team USA at the North American, Central American, Caribbean (NACAC) Cross Country Championships March 17 in Port of Spain, Trinidad.

In the women?s race, Huddle, the American 5,000-meter record holder, leads a field that includes Pan American Games gold medalist in the steeple chase Sara Hall (Flagstaff, Ariz.)(Montgomery HS, Santa Rosa 2001; Stanford 2005); five-time World Cross Country team member Renee Metivier Baillie (Boulder, Colo.); and 2011 Team USA steeplechaser Stephanie Garcia (Charlottesville, Va.).

On the men?s side, 2010 USA National Club Cross Country champion Braun will face 2011 USA 8 km champion Bobby Mack (Raleigh, N.C.) and 2008 NCAA Division I steeplechase champion Kyle Alcorn (Mesa, Ariz.)(Buchanan HS, Clovis 2003).

The championships will consist of six cross country championship races for men?s and women?s categories with national titles on the line for junior, masters and open divisions. The nation?s premier cross country event, the Championships will feature $35,000 in prize money.

Junior men
The junior men's race will feature Ohio State freshman Michael Bradjic, who competed for Team USA at last month's Great Edinburgh International Cross Country Team Challenge, where he finished seventh to help lead the squad to the team title.

Other contenders to watch include California state cross country champion Darren Fahy of La Costa Canyon HS in Carlsbad, along with Craig Nowak of Cypress Woods HS in Cypress, Texas, and Zach Herriott of Rockhurst HS in Kansas City. Nowak was third in the Nike Cross Nationals, and Herriott was ninth in the same race.

Junior women
Some of the leading contenders in the junior women's race include Sarah Fakler of Xavier College Prep in Phoenix, who competed for Team USA at last year's NACAC Cross Country Championships in Trinidad, and Grace Tinkey of First Presbyterian Day School in Macon, Georgia, who was sixth in the 2011 Foot Locker cross country championships in San Diego.

Julie Nacouzi (Montgomery HS, Santa Rosa 2011), who was a member of Team USA at last year's IAAF world cross country championships in Punta Umbria, Spain, is also in the field and could contend for the national title. Others to watch include University of Wisconsin freshman Jenna McMiller, and NCAA cross country championships competitor Shaylyn Tuite of Syracuse University.

Masters Championships
National championships will also be decided for masters men and women at 8 km with titles on the line in five-year age groups beginning at age 40 and up.

The fields will feature masters aces Tracy Lokken (Marquette, Mich.); and Kathryn Martin (Northport, N.Y.).
